Our study on DFCP1 and lysosome membrane repair is live! We uncover a previously unrecognized role for omegasomes in the cellular response to lysosomal membrane damage, revealing another layer of regulation in how damaged lysosomes are repaired. www.nature.com/articles/s41...

nature.comDFCP1-containing ER microdomains mediate lysosomal membrane repair - Nature Cell BiologyRadulovic et al. show that phosphatidylinositol 3-phosphate (PI3P), produced by the phosphatidylinositol 3-kinase PIK3C3, is formed on ER domains after lysosome damage. ER domain PI3P helps to recruit...

Yang et al. show that the secretory pathway kinase Fam20C is retained in the #Golgi via Fam20A-mediated membrane anchoring. Mislocalizing Fam20C alters #secretome phosphorylation, leading to reduced milk quality and subsequent growth retardation in pups. rupress.org/jcb/article/...

3/4 But why does high mTORC1 promote cell death under these conditions? High mTORC1 induces an anabolic state. However, when ER export is blocked, mitochondria become dysfunctional and cannot meet the metabolic demand. This energetic imbalance results in lower translation (despite high mTORC1 !!)

4/4 Our work highlights ER proteostasis as a therapeutic vulnerability in myeloma. This extends our previous work showing that targeting LTK, a kinase that regulates ER export, kills myeloma cells (www.nature.com/articles/s41...) We propose ER export as an avenue for multiple myeloma therapy

2/4 We show that targeting the ER export machinery induces death of MM cells. Surprisingly, cell death was unrelated to ER stress. Instead, we uncover this cascade: ER overload > activation of ERAD > proteasomal degradation > increase of free cytosolic amino acids > activation of mTORC1.

ERC success rates are tanking and the move is... even bigger grants for fewer people? smh

Katherine Stiles@katherinestiles.org

· #AcademicSky · The European Research Council is launching €7m Plus Grants for ambitious, long‑term research — open to researchers at any career stage but limited to about 30 awards; current ERC grantees and applicants are ineligible, reports @clanicholson.bsky.social.

Check out this new Roadmap, a fantastic team effort by a consortium of ER researchers. Glad we could provide the space for this synthesis, which came out of discussions at a fruitful meeting, see below:

It was a great pleasure to work on this paper. What started as a small meeting in January this year resulted in this beautiful piece of work.

Beautiful work which supports a role for the ER in mechanobiology.

💫NEW: Das, @fabianspill.bsky.social & co combine intracellular cartography and biophysical modelling to reveal how edge curvature governs endoplasmic reticulum (ER) morphology, showing that curvature-dependent ER reorganization directs distinct modes of epithelial cell migration. bit.ly/3HFAQ8S

Great preprint showing that COPII vesicles do exist in vicinity of ERES. Very interesting.

Liz Miller@lizmillercu.bsky.social

Go check out the latest preprint from Giulia Zanetti's lab. @dr-downes.bsky.social used cryo-tomography to directly visualize COPI and COPII coated vesicles in situ at unprecedented resolution in human cells. Amazingly beautiful, rigorous, insightful work. www.biorxiv.org/content/10.1...

ER needs ATP for processes like protein quality control, but can't make its own. Here, how ATP is imported into the ER lumen and exchanged for ADP.

Excited to present our unpublished data tomorrow on how Mechanosesing at the Endoplasmic Reticulum regulates Proteostasis. #Organelles #Mechanobiology
