[1/3] It was a lot of fun to work on optical simulations for real-world use cases of a K-mirror (image rotator) in two-photon microscopes with Shinjini Gosh, @ptrrupprecht.bsky.social, and the team. There are a few things that I learned on the way.

This is very solid work. It prompts a question I have never had a satisfying answer for: why do people want to infer spikes from calcium imaging data? The calcium signal contains sufficient information for *any* inference procedure, and none of the bias. Why don't people work directly with calcium?

Excited to share our work now in Nature Methods! We wrote this manuscript to help others better analyze and interpret calcium imaging data. As a central aspect, we show why nonlinear behavior of calcium indicators matters in practice. Very happy about this work!

Looking forward to #FENS2026 next week in Barcelona! I will present a poster on effects of locus coeruleus and arousal on astrocytes and neurons in hippocampus: PS06-09PM-201 PhD student Shinjini Ghosh will present a poster on a Theremin-based auditory virtual environment for mice: PS04-08PM-568

The @ptrrupprecht.bsky.social lab uses a really cool approach combining neuromodulator imaging, optogenetics, and two-photon calcium imaging to determine how locus coeruleus-mediated norepinephrine signaling influences different hippocampal cell types, including #astrocytes.

Excited to share our new preprint! We explore the link between the locus coeruleus (LC) and arousal for astrocytes, pyramidal cells, interneurons in the hippocampus. A fantastic collaboration with @sianduss.bsky.social @bohaceklab.bsky.social, and many others: www.biorxiv.org/content/10.6... 1/5

I honestly hate watching talks made with AI. I hate it when speakers brag about how much time it saved them. If you have 80 people in your audience and they all watch you for 30 minutes, that's 40 hours of person-time. And you're telling us you're *proud* of how little you prepared? 🤷‍♀️

Two brain circuits. Same learning problem. Shared learning outcomes. Different dynamical implementations. Our work shows that learning is not a single canonical solution but can emerge through distinct population dynamics shaped by circuit architecture. Excited to share our latest preprint! 🧵1/12

My review focuses on a point where data and interpretations across labs seem to diverge: the prevalence and importance of dendritic calcium and voltage events decoupled from the soma. I'd be interested in hearing more opinions on that!

1/8. New preprint! Using fUSi in head-fixed mice🐭, we found that arousal events trigger a brain-wide wave of activity 🌊🧠. Surprisingly, this pattern was preserved during opto manipulations of the locus coeruleus, pointing to a minor role for noradrenergic tone. www.biorxiv.org/content/10.6...

First time patching in months, y'all know what that means: I am about to spend the next few hours discovering fun and novel ways in which an ephys setup can break!

But then where do the branch-specific variations in voltage come from? We found that in apical dendrites, small subthreshold depolarizations *preceding* a complex spike partially predict voltage (and Ca2+) amplitude *during* the event. This suggests conjunctive amplification of inputs and spikes.
