Commentary: Christian Jorgensen @jorlab123.bsky.social discusses new study from Elhanafy et al. (https://buff.ly/4haX9PQ) which shows how identical mutations in the voltage sensing domain of sodium channels can yield differential functional effects. https://buff.ly/4jpCRUD#IonChannels
Nociceptor sodium #ionchannels shape subthreshold phase, upstroke, and shoulder of action potentials | Journal of General Physiology @JGenPhysiol https://buff.ly/4hku7gH#openaccess
1/5 Are you interested in the role of Nav1.9 in AP generation? And what do all the other #sodiumchannels do in the nociceptor? In a study led by the MD student Phil Köster in the lab, we performed manual patch clamp on all Navs known to be expressed in sensory neurons. Nav1.1,2,3,5,6,7,8,9.
Köster, Stiehl, @lampertlab.bsky.social and colleagues characterize eight sodium channel subtypes relevant to #nociception and use a computer model to simulate the response of two types of nociceptive nerve fibers. https://buff.ly/4hku7gH#Biophysics#Neuroscience#IonChannels
4/5 We model a CMi-fiber (sleeping nociceptor) and an Adelta-fiber. In both, Nav1.9 significantly contributed to the fast upstroke of the AP and also to the shoulder formation. We can also nicely mimic Nav related disease pathology, like erythromelalgia, in the model.
5/5 When stepwise reducing Nav1.9, we see the shoulder disappear. It fits to the observation that CMi-fibers have broad APs doi.org/10.1101/2024... and high expression of Nav1.9. So, the shoulder of nociceptor APs may well be supported not only by Nav1.8 but also - suprisingly strongly - by Nav1.9.

2/5 We find that – not surprisingly – Nav1.8 activates and inactivates at more depolarized potentials compared to all the other Navs. Nav1.9 – recorded by Enrico Leipold – and Nav1.5 activate at the most hyperpolarized potentials. Nav1.9 has hugh window currents, and prominent persistent currents.
3/5 What is the role of all those Navs during AP generation? Thomas Stiehl created a computer model in which we fed HH models of Navs, generated on our patch data, according to their expression revealed by transcriptomic data published by @dianatavf.bsky.social@tedpricethepainguy.bsky.social

How could simulations elucidate Nav1.5 #ionchannel blockers mechanism? | Journal of General Physiology | Rockefeller University Press https://buff.ly/4gTs9Em
Cool paper (and great resource). I’m especially keen the disconnect between mRNA and protein levels in adult tissues! Pay attention #devbiol, it’ll be the same in embryonic tissues.

5,562 proteins were found to have tissue-specific enrichment across 32 human tissues.
The majority of them (3,088) are enriched at the protein but not RNA level.
The brain hosts the largest number of proteins that are only enriched at the protein level but not RNA level.
Preprint alert: Using human microneurography protocols in PatchSeq experiments on pig DRGs we identify the molecular identity of human dermal CMi-fibers (sleeping nociceptors). We provide an integrated pig-human-more transcriptomic atlas and identify OSMR and SST as marker, backed by human expts.
OK, the named first author - Jannis Körner - even though without bluesky handle - needs a big shout out. He mastered the PatchSeq experiments at @rwth.bsky.social in the wonderful collaboration with Shreejoy Tripathy and Derek Howard at Toronto.
